• Recognizes human ADAM12
• ADAM12 (MLTN and Meltrin-alpha) is involved in skeletal muscle regeneration/ onset of cell fusion, macrophage-derived giant cells (MGC) and osteoclast formation from mononuclear precursors
  • Expressed in human malignant tumors
  • Involved in regulation of growth factor activities and integrin functions
  • ADAM12 can degrade extracellular matrix components likely allows it to detach cancer cells from basement membrane and assist on route to metastasis

Process:

• Antibody specific for ADAM12 pre-coated onto microwells
• ADAM12 protein in samples captured by the coated antibody after incubation
• After extensive washing, another antibody specific for ADAM12 is added to detect captured ADAM12 protein
• For signal development, horseradish peroxidase (HRP)-conjugated antibody is added, followed by Tetramethyl-benzidine (TMB) reagent
• Solution containing sulfuric acid used to stop color development and color intensity
  • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
