• Apoloprotein B (APOB) is a plasma protein synthesizes primarily in the liver
  • Essential for assembly of VLDL in liver and intestine

  • APOB gene encodes two isoproteins through mRNA editing (APOB-48 and APOB-100)
    • APOB-100: synthesizes by liver and essential for assembly of VLDL in liver

Process:

• Antibody specific for APOB pre-coated onto microwells
• APOB protein in samples captured by the coated antibody after incubation
• After extensive washing, another antibody of biotinylated specific for human APOB is added to detect captured human APOB protein
• For signal development, Streptavidin-HRP is added, followed by Tetramethyl-benzidine (TMB) reagent
• Solution containing sulfuric acid used to stop color development and color intensity
  • Proportional to quantity of bound protein, measurable at 450 nm with correction wavelength set at 630 nm
