Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human IGF-II. Anti-Human IGF-II specific antibody was purified by affinity chromatography employing an immobilized Human IGF-II matrix. Sandwich ELISA: To detect Human IGF-II by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Human IGF-II (500-P12Bt) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Human IGF-II. Western Blot: To detect Human IGF-II by Western Blot analysis this antibody can be used at a concentration of 0.1- 0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant Human IGF-II is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
The IGFs are mitogenic, polypeptide growth factors that stimulate the proliferation and survival of various cell types, including muscle, bone, and cartilage tissue in vitro. IGFs are predominantly produced by the liver, although a variety of tissues produce the IGFs at distinctive times. The IGFs belong to the Insulin gene family, which also contains insulin and relaxin. The IGFs are similar to insulin by structure and function, but have a much higher growth-promoting activity than insulin. IGF-II expression is influenced by placenta lactogen, while IGF-I expression is regulated by growth hormone. Both IGF-I and IGF-II signal through the tyrosine kinase type I receptor (IGF-IR), but IGF-II can also signal through the IGF-II/Mannose-6-phosphate receptor. Mature IGFs are generated by proteolytic processing of inactive precursor proteins, which contain N-terminal and C-terminal propeptide regions.Specifications
