Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Rat IL-10. Anti-Rat IL-10-specific antibody was purified by affinity chromatography employing an immobilized Rat IL-10 matrix. Sandwich ELISA: To detect Rat IL-10 by sandwich ELISA (using 100 μL/well antibody solution) a concentration of 0.5-2.0 μg/mL of this antibody is required. This antigen affinity purified antibody, in conjunction with PeproTech Biotinylated Anti-Rat IL-10 (500-P139Bt) as a detection antibody, allows the detection of at least 0.2-0.4 ng/well of Recombinant Rat IL-10. Western Blot: To detect Rat IL-10 by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 μg/mL. Used in conjunction with compatible secondary reagents the detection limit for Recombinant Rat IL-10 is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.
IL-10 encodes a protein that acts as a cytokine and is primarily produced by monocytes, with some production by lymphocytes. This cytokine has various effects on immunoregulation and inflammation, including down-regulating the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. Additionally, IL-10 can block NF-kappa B activity and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice have suggested that this cytokine is an essential immunoregulator in the intestinal tract. Mutations in this gene have been associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis. Diseases associated with IL10 include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.Specifications
