Description
MultiShot FlexPlate DH5α T1R Chemically Competent E. coli cells are cloning competent cells derived from a well-known, versatile DH5α strain that can be used in many everyday cloning applications. These cells are pre-aliquoted in a 96-well PCR plate to increase transformation productivity, and the FlexPlate format can be divided down to any combination of 8-well segments for flexibility in transformation throughput.
All of our T1R strains have the tonA (also known as fhuA) deletion in their genotype that confers resistance to T1 and T5 phage. T1 bacteriophage spread rapidly and lyse E. coli hosts, which are commonly used for cloning and library construction. A resistant genotype offers added security to protect valuable clones and libraries.
Reliable DH5α T1R cloning capabilities
MultiShot FlexPlate DH5α T1R competent E. coli cells are able to achieve transformation efficiencies of >1 x 108 cfu/μg control DNA. This strain is recommended for transformations of cloned unmethylated DNA, routine cloning of genes into large plasmids, and is suitable for difficult clone constructions, such as blunt-end ligations or cloning using limited amount of DNA.
The DH5α T1R strain carries the hsdR mutation to prevent restriction of foreign DNA and the endA1 mutation that increases the yield of plasmid DNA prepared using miniprep protocols. The lacZΔM15 marker provides α-complementation of the ß-galactosidase gene allowing blue-white screening on agar plates containing X-Gal or Bluo-Gal.
MultiShot FlexPlate DH5α T1R Competent Cells offer:
• Transformation efficiency of >1 x 108 transformants/μg plasmid DNA
• tonA (fhuA) genotype to confer resistance to T1 and T5 phage
• hsdR for efficient transformation of unmethylated DNA from PCR amplifications
• lacZΔM15 marker for blue-white screening
• endA1 for cleaner preparations of DNA and better results in downstream applications due to the elimination of non-specific digestion by endonuclease I
• recA1 for reduced occurrence of non-specific recombination in cloned DNA
Increase transformation productivity with MultiShot FlexPlate format
The FlexPlate is a 96-well PCR plate that can be used in automated liquid handling systems or divided into 8-well segments for flexibility in transformation throughput. Addition of DNA to the MultiShot FlexPlate Competent Cells can be quickly performed with a multichannel pipet or a liquid handling system. After addition of DNA, the cells can be efficiently transformed by heat-shock using a water bath, heat block, or thermal cycler.
Key benefits of the MultiShot FlexPlate format
• Increased throughput—compatible with automated liquid handling platforms
• Flexible—transform in increments of 8 wells to entire 96-well plate
• Convenient—competent cells pre-aliquoted into 96-well plate, ready for transformation
