Description

The Human Amyloid Beta 42 (Hu Abeta 42) ELISA quantitates Hu Abeta 42 in human tissue culture medium, tissue homogenate, and cerebrospinal fluid (CSF). The assay will exclusively recognize both natural and recombinant Hu Abeta 42. Principle of the method The Human Abeta 42 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Rigorous validation Each manufactured lot of this ELISA kit is quality tested for criteria such as sensitivity, specificity, precision, and lot-to-lot consistency. Easy-to-run sandwich ELISA
The Human Aβ42 kit is a solid-phase sandwich enzyme-linked immuno-sorbent assay (ELISA). A monoclonal antibody specific for the NH2-terminus region of human Aβ has been coated onto the wells of the microtiter strips provided. During the first incubation, standards, controls, and unknown samples are pipetted into the wells and co-incubated with a rabbit antibody specific for the COOH-terminus of the 1-42 Aβ sequence. Bound rabbit antibody is detected through the use of a horseradish peroxidase-labeled anti-rabbit antibody. After washing, horseradish peroxidase-labeled anti-rabbit antibody (enzyme) is added. After a second incubation and washing to remove all unbound enzyme, a substrate solution is added, which is acted upon by the bound enzyme to produce color. The intensity of this colored product is directly proportional to the concentration of human Aβ42 present in the original specimen. Samples are quantitated using a standard curve generated from the recombinant Aβ42 included with the kit.

The kit comes with the capture antibody pre-coated to the wells, eliminating the need to coat the plate. No overnight incubation needed. The incubation steps can be performed in four hours.

Large dynamic range: 7.1–1,000 pg/mL
The large dynamic range of the assay minimizes the need to guess the dilution needed for your sample to fall within the linear assay range.

Economical, removable wells
The precoated wells come as removable 8-well strips, allowing assay size to tailored to needs. Two standard vials per plate provided allowing the option of reconstituting a new vial should if the entire plate is not run at once.
