Description
Concentrations of albumin and IgG can make up more than 70% of total serum proteins and often mask lower quantity proteins during protein-profiling experiments. Optimize your serum sample prep by removing albumin or IgG quickly and conveniently. By using our depletion kits to remove excess high-abundance serum proteins, less abundant proteins of interest can be easily detected by 2D electrophoresis and other protein profiling methods such as mass spectrometry.
Pierce Albumin/IgG Removal Kit• Optimized—microcentrifuge spin cup method is scaled for treating 10 μL human serum samples for 2D electrophoresis
• Efficient—removes nearly all HSA and IgG, enabling low-abundance proteins to be detected in 2D gels
• Fast—processes samples in less than 40 minutes
• Scalable—use different amounts of affinity resin to match sample volume and concentration
The Thermo Scientific Pierce Albumin/IgG Removal Kit uses a classical mixture of Cibacron Blue Dye and Protein A resin for removal of albumin and IgG. The dye binds to albumin with good specificity, and Protein A binds to many species and subclasses of IgG. The Pierce Albumin/IgG Removal Kit is economical and can be used to treat samples from other species besides human (e.g., monkey, swine, rabbit). The kit is also scalable and sufficient to process 25 samples, each containing 600 μg of total serum protein (about 10 μL of serum).
Pierce Albumin Depletion Kit
• Complete—includes optimized buffer and microcentrifuge spin columns to remove albumin quickly and conveniently from 10- to 50-μL samples
• Adaptable—easy-to-dispense slurry improves performance, eases processing of multiple samples, and can be adapted to larger or smaller columns or 96-well filter plates
• Compatible—removing excess albumin facilitates MS analysis or electrophoresis gel analysis of low-abundance serum proteins
The Thermo Scientific Pierce Albumin Depletion Kit resin is supplied as a 50% slurry. To prepare the depletion columns, 200 μL of slurry is dispensed into the supplied microcentrifuge spin columns to obtain 100 μL of settled beads to process 10 to 50 μL of serum sample in a single reaction. The kit has been optimized to bind human, porcine, and sheep albumin from serum samples. With a modification to the protocol, albumin from bovine, calf, goat, and rat can be removed with this method. The kit is not effective for removal of mouse albumin. Processed samples are ready for immediate downstream analysis by electrophoresis or MS applications.
